Persistent organic pollutants (POPs) are a broad group of compounds with varying physicochemical and toxic properties. Among POP, polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) are two families of hydrophobic compounds sharing some similar environmental properties such as hydrophobicity, persistence, bioaccumulation and toxicity. Previous studies have demonstrated that these compounds could affect fish life-history traits such as survival, growth and reproductive success. Étude des voies de bioactivation du benzo[a]pyrène et du fluoranthène chez la sole commune (Solea solea) : Profil métabolique et génotoxicité ArchiMer

The aim of this Ph‟D was to study the biotransformation and the genotoxicity of two PAH of interest, namely the benzo[a]pyrene (BaP) and the fluoranthene (Fluo), in the common sole Solea solea, which is a benthic species sensitive to chemical pollution. The use of in vitro exposure models and the application of two different chemical analytical techniques allowed demonstrating that, as in higher vertebrates, the dihydrodiols and the phenols of BaP and Fluo are the major metabolites produced by sole liver enzymes. In vitro, the genotoxic potential of BaP and Fluo was demonstrated in sole, by the production of DNA adducts and oxidative DNA damage. Le mercure en milieu marin. Biogéochimie et écotoxicologie ArchiMer

This report synthesizes the knowledge on the biogeochemical cycle, bioconcentration processes and ecotoxicity of mercury in the marine environment. Man induced changes on the mercury cycle, especially along the French coasts are reviewed. Concentration levels, mechanisms and fluxes between geochemical reservoirs are assessed. Emphasis is given on sublethal effects and toxicity mechanisms. Quality standards used in European Community countries are listed. Conclusions and recommendations for environmental management and research are given.

The oyster Crassostrea gigas is one of the most commercialized molluscs in the world. It is a very cosmopolitan species that has been disseminated by humans and is susceptible to various agents such as pathogens (bacteria, viruses), parasites, xenobiotics and/or toxic micro-algae. As a filter-feeder, it may accumulate phyco- toxins while feeding on harmful algal blooms (HAB), including paralytic shellfish toxins (PST). To protect human health shellfisheries harvesting are closed dur- ing HAB, impacting the aquaculture industry. The originality of this project consists in the interdisciplinary approach gathering the impact of toxic phytoplankton blooms and metallic pollution on health (behaviour, stress, immunity, genetic) and disease development of the Manila clam, an economieally important species. The Manila clam Ruditapes philippinarum is the second exploited bivalve in the world. It was introduced in Arcachon bay during the 80's and is today exploited by professional fishermen (57 licences). A 50% decrease of the stock was observed during the last five years. Test écotoxicologique sur microalgues et lecteur de plaques : mesures d'activités enzymatiques sur cellules in vivo. Application à l'évaluation de la qualité du milieu ArchiMer

A new method for the assessment of several contaminant effects on marine microalgae (Tetraselmis suesica, Skeletonema costatum) has been developped. The method is based on the measure of cells esterase activity using a fluorimetric stain, fluorescein diacetate (FDA). The experiments are performed in microplates using a fluorimetric microplate reader. The different steps of the experiments and the data processing are controlled by an original specific software. Application of the system for rapid ecotoxicological tests (determination of LC 50) and for assessment of environment quality by observation of water samples are proposed.

Utilisation des biomarqueurs pour la caractérisation de l’état écotoxicologique des masses d’eau ArchiMer

In the past 25 years, numerous biomarkers have been developed with the objective to apply them to aquatic ecosystem biomonitoring. Recently, the Water Framework Directive (WFD) of the European union specified monitoring programs required to assess the achievement of good chemical and ecological status for all water bodies by 2015. This article reviews the potential of biomarkers for ecotoxicological status assessment in WFD monitoring programmes based on previous national and international experiments.
